适应症
本品用于单纯饮食控制和或运动疗法血糖水平未得到满意控制的2型糖尿病病人，可作为单用磺脲类或盐酸二甲双胍治疗，血糖水平未得到满意控制的2型糖尿病病人二线用药。
用法用量
个性化给药。 1.饮食与运动疗法已不能满意控制血糖的2型糖尿病患者，推荐开始剂量为1日1次，1次1片(格列本脲/二甲双胍:1.25mg/250mg)，与饭同服。 2.对于基线HbAlc>9%或FPG>200mg/dl的初次治疗病人，开始剂量为1日2次，1次1片(1.25mg/250mg)，早晚饭时服。每隔两周，可增加本品的每日剂量，以达到最小的有效治疗剂量。 3.用于以前已服用过其它降糖药的病人，推荐开始剂量为1日2次，1次2片(2.5mg/500mg)或1日2次，1次4片(5mg/1000mg)，与饭同服。 4.对于单用磺脲类或盐酸二甲双胍已不能很好控制血糖的病人，推荐开始剂量为1日2次，1次2片(2.5mg/500mg)或1日2次，1次4片(5mg/1000mg)，与早晚饭同服，为避免低血糖，开始剂量不应超过以前所服的磺脲类或盐酸二甲双胍的日剂量，日剂量逐步调整，每次增加量不超过2片(2.5mg/500mg)，直达到最小的有效治疗剂量，建议最大日剂量不超过8片(10mg/2000mg)。
不良反应
骨髓抑制，间质性肺炎及肺纤维症，恶心，呕吐，腹泻，头晕，皮疹等。
禁忌
下列患者禁用:
1.肾脏疾病或者肾功能不全(男性:血清肌酐≥1.5mg/dl;女性:血清肌酐≥1.4mg/dl。或肌酐清除率异常)，由心源性休克、心肌梗塞、败血症引起的肾功能不全。
2.需药物治疗的充血性心衰。
3.对二甲双胍或格列本脲过敏者。
4.急慢性代谢性酸中毒，包括糖尿病性的酮症酸中毒。
5.静脉肾盂造影或动脉造影前、重大手术以及临床有低血压和缺氧情况。
6.Ι型糖尿病患者。
注意事项
1.低血糖：本品可能引起低血糖，当热量摄取不足、剧烈运动没有及时补充热量，同时使用其它降糖药或乙醇时发生低血糖的危险增加，肾或肝功能不全可升高格列本脲和盐酸二甲双胍的血药浓度，而肝肾功能不全可能减弱糖异生作。所有这些都增加低血糖的危险性。
2.服药期间患者应经常检查肾功能、肝功能、血糖，并进行眼科检查等，盐酸二甲双胍主要经肾脏排出，盐酸二甲双胍积聚和乳酸酸中毒的危险性随着肾功能损害的程度而增。
3.慎用影响肾功能或二甲双胍消除的药物。
4.避免过量饮酒。
5.肝功能不全者不宜用。
6.适量补充体内VitB12。
7.2型糖尿病患者在服用期间发现一些临床疾病（特别是迷走神经疾病）应立即检查是否是酮症酸中毒或乳酸酸中毒，此时应评估血浆电解质酮类，血糖水平是否正常，若出现血浆pH值下降或血中乳酸盐、丙酮酸盐和二甲双胍水平升高，立即停止服用，并采用其它手段进行纠正。
